During the inflammatory process, the
activation of phospholipase A2 will
induce the enzymatic cascade of
arachidonic acid, leading to the
release of prostaglandins which are
the main inflammation mediators.
Mechanism of action
Desonide induces the expression of
lipocortin which inhibits the
phospholipase A2 blocking the
release of arachidonic acid. This will
result in a decreased production of
prostaglandins reducing all the
inflammation signs.
The phosphate formulation ensures a long surface activity. At
corneal level the phosphate form comes in contact with phosphatase
enzymes and the desonide is converted in its alcoholic form: only
after this conversion the steroid molecule will be able to penetrate the
cornea. This converting process provides a long persistence
at ocular surface level.
Because of the lower deep penetration, the side effects are reduced resulting in a higher safety.
